pRS416Tef1-Ura3
PLASMIDThe plasmid for PCR amplification of the promoter replacement cassette with the URA3 selectable marker was constructed by amplifying the loxp-URA3-loxp region from pUG72 (Gueldener et al., 2002) with primers S19 and S20. The amplicon was placed 5′ of the translation elongation factor-1a (TEF) promoter region in p416Tef using homologous recombination in yeast.
