STRAIN

EHYL10 (JBx_055413), pGPDIN-fadB and pFBAIN-aco were integrated with linearized pEHYl087 (JBx_056155). The Yarrowia selection marker was excised. The E. coli fadB (EcDH1_4135) and the M. luteus aco (Mlut_11700) were codon-optimized for expression in S. cerevisiae. BsaI and BsmBI restriction sites were removed to allow for Golden Gate assembly. A three amino acid long Yarrowia peroxisome targeting signal (SKL) was added to both the fadB and aco carboxy-terminus.
