STRAIN

DH1 was sequentially transformed with pKD46 followed by pTE50; cells were made competent in the presence of ~25uM arabinose; a Hphmx6 cassette with flanking homology to the cydC locus was then electroporated into cells and after outgrowth plated on LB Hyg70 Kan plates. Candidate integrants were validated for on-target integration at the cydC locus using outside-outside primers specific to the genomic locus and absent from pTE50.. . Spontaneous SucR resistant colonies will arise roughly 1 in every ~50,000 CFUs as cydC is an essential gene.
